چکیده

reduced contractile function. Objective: To unravel mechanisms underlying reduced sarcoplasmic reticulum (SR) Ca release in persistent AF. Methods: We studied cell shortening, membrane currents, and [Ca ]i in right atrial myocytes isolated from sheep with persistent AF (duration 129 39 days, N 16), compared to matched control animals (N 21). T-tubule density, ryanodine receptor (RyR) distribution, and local [Ca ]i transients were examined in confocal imaging. Results: Myocyte shortening and underlying [Ca ]i transients were profoundly reduced in AF (by 54.8% and 62%, P<0.01). This reduced cell shortening could be corrected by increasing [Ca ]i. SR Ca 2 content was not different. Calculated fractional SR Ca release was reduced in AF (by 20.6%, P<0.05). Peak Ca current

Ultrastructural and functional remodeling of the coupling between Ca2+ influx and sarcoplasmic reticulum Ca2+ release in right atrial myocytes from experimental persistent atrial fibrillation.

RATIONALE Persistent atrial fibrillation (AF) has been associated with structural and electric remodeling and reduced contractile function. OBJECTIVE To unravel mechanisms underlying reduced sarcoplasmic reticulum (SR) Ca(2+) release in persistent AF. METHODS We studied cell shortening, membrane currents, and [Ca(2+)](i) in right atrial myocytes isolated from sheep with persistent AF (durat...

Impact of Sarcoplasmic Reticulum Calcium Release on Calcium Dynamics and Action Potential Morphology in Human Atrial Myocytes: A Computational Study

Electrophysiological studies of the human heart face the fundamental challenge that experimental data can be acquired only from patients with underlying heart disease. Regarding human atria, there exist sizable gaps in the understanding of the functional role of cellular Ca²+ dynamics, which differ crucially from that of ventricular cells, in the modulation of excitation-contraction coupling. A...

New manifestations of electrophysiological remodeling of heart during experimental model of atrial fibrillation in cirrhotic rat isolated heart

Introduction: The present study is aimed to evaluate electrophysiological remodeling of atrioventricular (AV) node and ventricular conduction during experimental atrial fibrillation (AF) model in isolated heart of cirrhotic rats. Methods: Cirrhosis-induced electrophysiological remodeling was evaluated in 24 isolated retrogradely perfused rat hearts in 2 groups (control and cirrhotic). Cirrho...

Enhanced sarcoplasmic reticulum Ca2+ leak and increased Na+-Ca2+ exchanger function underlie delayed afterdepolarizations in patients with chronic atrial fibrillation.

BACKGROUND Delayed afterdepolarizations (DADs) carried by Na(+)-Ca(2+)-exchange current (I(NCX)) in response to sarcoplasmic reticulum (SR) Ca(2+) leak can promote atrial fibrillation (AF). The mechanisms leading to delayed afterdepolarizations in AF patients have not been defined. METHODS AND RESULTS Protein levels (Western blot), membrane currents and action potentials (patch clamp), and [C...
